Help needed with Permutation and Combinations

To start off, I understand the basic concept of permutation and combinations.
This is my dilemma, I am in the midst of writing an essay about mathematics and I chose to do an evaluation on the possibilities of four letter words (26^4) and the amount that are actually used. I am dwindling down the amount of words possible by looking at certain 'rules' of the English language such as each word must have at least 1 vowel and no real 4 letter has a repetition of a letter three or four times (like aaaa or aaab).
I wasn't sure if there was easy way to make these calculations or if I have to make long calculations for each instance.
